The city is Estrie's major Institutional, political, cultural and economic centre. At the start of the 20th century, the city was additionally referred to as the Queen of the Eastern Townships.

The city of Sherbrooke is a crucial university centre. The city of Sherbrooke has 8 institutions which employ 11,000 people, 3700 of which are teachers, researchers and professors. There are more than 40,000 students at the university. The direct economic effect of all of these institutions exceeds over one billion dollars. Since the 19th century, Sherbrooke has been an important manufacturing center. American Loyalists were the initial inhabitants of the city in the year 1793. Part of the area has been historically referred to as the Eastern Townships. Farmer Gilbert Hyatt was initially from Schenectady, New York. During 1802, he constructed a flour mill. The community was called "Hyatt's Mills" until the year 1818. At that time, the town was renamed after Governor General Sir John Sherbrooke, when he retired and returned to England. The vast majority of the residents within the city of Sherbrooke speak French, despite the township's Anglophone name and heritage; there are hardly any traces of Sherbrooke's English past remaining.

On January 1, the year 2002, the city grew significantly by the merges of the cities of Sherbrooke, Deauville, Ascot, Lennoxville, Bromptonville, Saint-Elie-d'Orford, Rock Forrest and Fleuimont.
